PhD Summary

The research project was initiated with the goal of developing a new application for the established trace gas analysis technique, selected ion flow tube mass spectrometry (SIFT-MS), viz. monitoring the headspace of large scale human cell cultures in real time; particularly those of importance to the field of regenerative medicine.
Some of the major activities that were undertaken as part of the project include analysis of the gas phase above various cultures of primary and immortalised human cell types using SIFT-MS,
culture of human mesenchymal stem cells and hepG2 hepatocarcinoma cells on microcarriers in spinner flasks bioreactors and a larger (1L) disposable bag-type bioreactor and the development of headspace/gas phase sampling methodologies for the analysis of emitted compounds.
A study of ethanol/acetaldehyde and DMSO metabolism by cultured human cell types. Aldehyde dehydrogenase inhibitors (dissolved in DMSO) were applied to the cells and the changes in acetaldehyde and dimethyl sulphide concentrations were detected via SIFT-MS analysis of the culture headspace.
Analysis of the gas/vapour compounds emitted by microbial cell cultures. Of particular interest is a study of the volatile compounds emitted into the gas phase by E. coli (JM109), when cultured in two different media. In this study, the real time capability of SIFT-MS was also utilised to monitor the production of these compounds in the gas phase above one such culture.

Current Employment

Currently conducting research at Keele University along two strands; one relating to the development of a cell counting method based on gas phase analysis of human cell cultures and the other on analysing the volatile compounds emitted into the headspace of cultured microbial cells that commonly infect the lungs of cystic fibrosis sufferers, as a part of a larger project aimed at using breath analysis to diagnose the presence of specific respiratory tract infections
